The polyQ region of the endogenous VAChT gene has been extended by one glutamine residue, to 14 residues. A loxP cassette containing a Disc\RFPDsRed(T1).3xP3.cHa marker has also been inserted into the genome as part of the mutagenesis procedure.

VAChT14Q homozygous late stage embryonic aCC/RP2 motoneurons exhibit significantly reduced amplitude and frequency of spontaneous mEPSC when compared to controls. Also they lack any observable evoked spontaneous rhythmic currents (SRCs) or optogenetically (using Crei\ChR2CHETA(E123T.H134R).UAS.YFP under the control of Scer\GAL4ChAT.BAC) evoked EPSCs different than controls.
