Description

Mobilization of the progenitor P{EPgy2}Pol32EY15283 insertion, resulting in deletion of nt15255502 to nt15256304 with an addition of 39bp of P element sequences. Since only part of the Pol32 coding region is removed, along with the entire annotated 3'UTR, there is still a possibility that a truncated Pol32 protein with 219 residues is produced, though this is undetectable by Western Blot analyses.

Comment:

An 803 bp deletion resulting from the imprecise excision of P{EPgy2}Pol32EY15283. Reported to map to 2L:15255502..15256304 . 39 bases of P element sequences remain at the site of the excision.

Detailed Description

Pol32L30 mutant adults express a variable degree of bristle loss or shortening, as compared to controls. Females are sterile, as they lay eggs that are fertilized but do not show more than 8 nuclei; in agreement, most of the DNA from these embryos is mitochondrial.

A small proportion of Pol32L30/Pol32L27 third instar larvae show double strand breaks, which are nearly absent in controls. A very small proportion of adults show an etched abdomen phenotype.

A small proportion of Pol32L30/Pol32L27 third instar larvae show double strand breaks, both centric and non-centric, which are nearly absent in controls; these become much more frequent in a DNApol-δI10 heterozygous background and are both centric and non-centric.

A very small proportion of Pol32L30/Pol32L27 adults show an etched abdomen phenotype; this phenotype is only slightly increased in a DNApol-α180G13925/+ background, is moderately increased in a Pol31G16501/+ background, becomes highly prevalent in Df(3L)BSC122/+ or Df(3R)Exel6186/+ backgrounds and becomes nearly fully penetrant in a DNApol-δI10/+ background; the phenotype is not much affected in Df(3R)X3F/+, spn-A1/+, Df(3R)P-21/+, or BlmD2/+ backgrounds.
