Single nucleotide change inducing the substitution of the glycine (at position 629 for orion isoform A and 611 for orion isoform B) into an aspartic acid. The mutation lies in the common shared part of the isoforms and therefore affects both orion-A and -B functions.

orion1 adults show a highly penetrant, cell non-autonomous mushroom body axon pruning phenotype, as revealed by the presence of adult unpruned vertical gamma axons as well as the strong presence of debris. The unpruned axon phenotype is particularly apparent during metamorphosis; no glial cell invasion of the gamma bundle is observed at 6h APF, and glial infiltration and engulfment of the degenerated larval axons is not observed at 12h APF nor at 24h APF. Dendrite pruning is also clearly affected.

orion1/Df(1)BSC622 adults also show a mushroom body axon pruning phenotype.
