UAS sequences (from pWalium10.moe) regulate expression of full-length, pathogenic Hsap\ATXN3 that encodes a polyglutamine (polyQ) repeat of 80 residues (the polyQ repeats are encoded by CAGCAA doublet repeats to preclude mRNA toxicity and unconventional translation). The coding sequence contains a K117R amino acid substitution (mutation of the primary ubiquitination site to a non-ubiquitinatable residue). The polyQ-reading frame is tagged at the C-terminal end with Tag:HA.
