20xUAS sequences regulate expression of a transcript that encodes the two components of the light-inducible microtubule disassembly system (MTDS) described in PMID:35686621, separated by a 2A linker (this results in separate translation of each protein). The first component, 'EMTB-mCherry-CIBN', consists of mCherry that is tagged at the N-terminal end with the Tag:MT(EMTB) microtubule localization sequence and at the C-terminal end with CIBN(ΔNLS). The second component, 'dNSpastin3Q-CRY2', consists of a truncated form of Mmus\Spast that includes the region sufficient for microtubule severing and in which 3 key residues required for microtubule-binding have each been mutated to Q, tagged at the C-terminal end with CRY2(PHR). The EMTB-mCherry-CIBN protein localizes to microtubules due to the presence of the Tag:MT(EMTB) tag. Upon illumination with blue light, the CIBN(ΔNLS) and CRY2(PHR) peptides dimerize, recruiting the dNSpastin3Q-CRY2 protein onto the microtubules, which are then rapidly disassembled.
