Overview

This report describes a model of epithelial cancer using the Drosophila syntaxin gene Syx7; there are two human genes orthologous to Dmel\Syx7 : STX7 and STX12. The syntaxin gene family encodes membrane receptors involved in vesicle transport as components of SNARE (SNAP receptor) complexes. Neither human gene has been implicated in cancer. Classical loss-of-functions mutations, RNAi-targeting constructs, and alleles caused by insertional mutagenesis have been generated for Dmel\Syx7.

Neither human gene, STX7 or STX12, has been introduced into flies.

Animals homozygous for an amorphic allele of Dmel\Syx7 die during the larval stage. Assayed in imaginal discs, somatic clones homozygous for an amorphic allele of Dmel\Syx7 are slow-growing and usually eliminated from the disc. Very large clones produced using the Minute system exhibit epithelial disorganization and undergo massive overproliferation during an extended larval stage.

Molecular information

The syntaxin gene family encodes membrane receptors involved in vesicle transport as components of SNARE (SNAP receptor) complexes. The STX7 protein mediates the endocytic trafficking from early endosomes to late endosomes and lysosomes. STX12 is a component of a SNARE complex that acts to regulate protein transport between late endosomes and the trans-Golgi network. [Gene Cards, STX7, STX12; 2019.02.12]

Note that STX13 has been determined to be the same gene as STX12.

Collins et al. (2002, pubmed:12218144) showed that STX7 and STX12 are involved in the ordered fusion of endosomes and lysosomes with the phagosome, where phagocytic cells kill and degrade internalized foreign particles. STX12 is localized to the recycling endosome compartment, while STX7 is found in late endosomes and lysosomes and both are recruited to the phagosome. However, STX12 is acquired earlier before rapidly recycling off the phagosome, while STX7 is recruited later and continues to accumulate throughout the phagosome maturation process.
