Third instar larvae expressing Hsc70-5KK100233 under the control of Scer\GAL4elav.PU are sluggish: their crawling velocity is reduced and the righting reflex delayed. Their neuromuscular junction exhibits defects: mitochondria are decreased in content (fewer, smaller and rounder mitochondria), increased proportion of postsynaptic densities unopposed by active zones, and microtubule defects (fewer microtubule loops in terminal boutons and fewer boutons connected to a stable microtubule network); there are more frequent spontaneous excitatory junctional potentials but with decreased amplitude and decreased quantal content, decreased evoked excitatory junctional potentials, and progressive increase in evoked excitatory junctional potential failure upon 10Hz stimulation.

Induced expression of Hsc70-5KK100233 under the combined control of Scer\GAL4elav.PU from 5d after egg laying allows individuals reaching adulthood. Adults exhibit decreased climbing ability and abnormal wing posture, and are short lived.

The expression of Hsc70-5KK100233 under the control of Scer\GAL4c564 leads to lethality.

Expression via Scer\GAL4tub.PU results in fragmented mitochondria and decreased mitochondrial membrane potential in peripodial cells of wing discs.

Expression via Scer\GAL4tub.PU results in increased mitophagy in peripodial cells of wing discs.

Expression via Scer\GAL4esg-NP5130 results in increased cell death of adult midgut progenitors.

Expression of Hsc70-5KK100233 under the control of Scer\GAL4GMR.PF does not cause any changes in external eye morphology upon fly ageing.

Knockdown of Hsc70-5 through expression of Hsc70-5KK100233 in DA neurons using Scer\GAL4Ddc.PL or Scer\GAL4ple.PF, but not in sensory neurons, results in lethality during the larval or pupal stages.

Knockdown of Hsc70-5 through expression of Hsc70-5KK100233 in motoneurons using Scer\GAL4RapGAP1-OK6, Scer\GAL4VGlut-OK371, or Scer\GAL4D42, results in either larval or pupal lethality.

Expression of Hsc70-5KK100233 in the developing eye, under the control of Scer\GAL4GMR.PF does not cause degeneration in the external eyes of adult flies.

Pan-neuronal expression of Hsc70-5KK100233 under the control of Scer\GAL4elav-C155, at 18[o]C, results in death before eclosion to adulthood. Larvae display no abnormal body-posture at rest or during locomotion.

Suppression of early lethality in flies expressing Hsc70-5KK100233 under the control of Scer\GAL4elav-C155, through co-expression of Scer\GAL80ts.αTub84B, results in locomotion and body posture defects. These flies exhibit a 50% decrease in cellular ATP levels.
