Recent work in human has implicated the gene RNF220 in a syndrome characterized by leukodystrophy with ataxia, deafness, liver dysfunction, and dilated cardiomyopathy (HLD23); HLD23 exhibits autosomal recessive inheritance. RNF220 encodes an E3 ubiquitin-protein ligase; there is a single orthologous gene in Drosophila, also designated RNF220. Neither the human gene nor the fly gene has been extensively characterized. A small number reagents have been generated for Dmel\RNF220 including RNAi-targeting constructs, overexpression constructs, and alleles caused by insertional mutagenesis.
The human RNF220 gene has not been introduced into flies.
Effects of RNAi-mediated knockdown of Dmel\RNF220 in larval muscle, the developing eye, and larval salivary glands have been investigated. Knockdown at 25 degrees is more effective than at 18 degrees, and results in adult lethality in most cases. Knockdown specifically in the developing eye at 18 degrees or later in development results in viable adults with severe neurodegeneration of the retina. Knockdown of RNF220 in the larval salivary glands results in significantly altered subcellular localization and aggregation of the fly lamin encoded by Dmel\Lam, but not of the product of Dmel\LamC. Results in Drosophila support findings of a functional link between RNF220 and lamin B1 in human.

Hypomyelinating leukodystrophies constitute a subset of genetic white matter disorders characterized by substantial and permanent deficits in CNS myelin deposition. Most patients with severe hypomyelination present in infancy or early childhood and develop severe neurological deficits; clinical presentation can also be mild with onset of symptoms in adolescence or adulthood (Wolf et al., 2021; pubmed:33324001).

AR-LAD is described as a leukodystrophy with ataxia and sensorineural deafness that includes fibrotic cardiomyopathy and hepatopathy as associated features (Sferra, et al, 2021, pubmed:33964137; FBrf0252013).
Leukodystrophies are a heterogeneous group of rare inherited disorders that mostly involve the white matter of the CNS. These conditions are characterized by primary glial cell and myelin sheath pathology of variable aetiology, which causes secondary axonal degeneration, generally emerging with disease progression. Hypomyelinating leukodystrophy 23 with ataxia, deafness, liver dysfunction, and dilated cardiomyopathy (HLD23) is an autosomal recessive neurodegenerative disorder with systemic manifestations. Affected individuals show delayed motor development and ataxic gait in early childhood that progresses to spastic paraplegia with loss of ambulation in the first decades of life. Additional features include progressive sensorineural hearing loss resulting in deafness, hepatic dysfunction with elevated liver enzymes, and dilated cardiomyopathy that ultimately results in death in the first or second decades. Two homozygous missense variants (p.R363Q and p.R365Q) in the ubiquitin E3 ligase RNF220 as the underlying cause of this syndromic form of leukodystrophy in seven consanguineous families; the disease exhibits autosomal recessive inheritance (Sferra, et al, 2021, pubmed:33964137; FBrf0252013).
