Comments concerning this interaction

Interaction in vitro; bait produced as a recombinant fusion protein in bacterial system; prey derived from transfected CHO cell extract.

nuf binds to Rab11 but not Rab5.

Binding of nuf to Rab11 is not tightly linked to the state of the nucleotide, as binding occurred in the presence of a GTPγS or GDPβS.

Source was fixed follicle cells of transgenic fly line; bait produced from tagged transgenic construct; prey produced from tagged transgenic construct.

Bait protein was apical. Prey protein, on its own, was localized throughout the cell on vesicle-like structures. The ability of bait protein to recruit prey protein to the apical cytoplasm was taken as evidence of direct physical interaction.

Comments concerning this interaction

Interaction in vitro; bait produced as a recombinant fusion protein in bacterial system; prey derived from S2 cell detergent-free extract (endogenous gene).

Specificity S-score=12.69.

Constitutively active (GTP-locked) form of Rab was used for pull down. Parallel pull downs performed for several Rabs proteins in this study. Yield was measured using spectral counts. A specificity score (S score) was calculated, where greater weight was given to interactions seen with fewer baits.

Comments concerning this interaction

Interaction in vitro; bait produced as a recombinant fusion protein in bacterial system; prey produced as a recombinant fusion protein in bacterial system.

Mutation of nuf\'s aPKC phosphosite (Ser155) did not affect binding to Rab11. Also, aPKC did not compete with Rab11 for binding to nuf.
