Dear FlyBase, My name is Toby Wu and I am an undergraduate working in Dr. Celeste Berg's laboratory at the University of Washington. Two years ago, I sequenced two alleles of Drosophila ras, Ras85DSutor-341 and Ras85DSutor-404, as part of my project. I have included the molecular data for these two alleles below. Ras85DSutor-341 consists of a nonsense mutation that affects amino acid \#131 (Arg to Stop). The insertion of a new stop codon 3bp upstream of the second intron results in truncation of the protein, thereby eliminating the CAAX box. Ras85DSutor-404 consists of a 5' splice site point mutation in intron 1, which changes the conserved GU to AU. The altered 5' splice site may no longer serve as a docking site for the U1snRNP complex of the spliceosome, and subsequently lead to incomplete RNA splicing.
