I'd like you to revisit P{hs-FLPD5.fco} (FBtp0093779) and the associated insertions P{hs-FLPD5.fco}attP3 (FBti0161061) and P{hs-FLPD5.fco}attP40 (FBti0161057).
Barret Pfeiffer, Aljoscha Nern, Gerry Rubin and colleagues generated a series of constructs expressing different versions of FLP recombinase. They found significant activity differences between FLP with glycine versus aspartic acid at position 5 and the constructs are listed in FlyBase with either "G5" or "D5" designations.
Barret Pfeiffer sent stocks to Bloomington with an unpublished construct where a FLP sequence had been codon-optimized for flies. FLP is expressed under the control of heat shock regulatory sequences and it carries the same PEST sequence present in constructs from Nern et al. (FBrf0214794). He sent it with the symbol pBPhsFlpPEST-Opt. It's currently listed in FlyBase as P{hs-FLPD5.fco} where "fco" was used to indicate "fly codon optimized". Unfortunately, I made a mistake in my correspondence with FlyBase (FBrf0225079) and said the construct encodes aspartic acid at position 5 (D5) when it really encodes glycine (G5) at that position. I found out about this mistake when David Bilder resequenced the construct and I reviewed my old correspondence with Barret.
We will withdraw the stocks from distribution under their current stock numbers and begin distributing them under new stock numbers with updated genotypes. I suggest you change the transgene symbol to P{hs-FLPG5.PEST.Opt} to match the original symbol more closely.
