Curator question - Based on the primer sequences, I have mapped the PtthDelta deletion to 2L:576323..576922 , a deletion of 600bp, but it was reported to be a 604bp deletion. Can you please send the corrected coordinates for the deleted region?
I have checked all the steps for the generation of the PtthDelta deletion. Indeed, all of the information in Shimell et al, 2018 (FBrf0238358) regarding the cloning procedure is correct. However, as you have noticed, the deletion contains only 600nt (and not 604nt). Unfortunately, the number of nucleotides was miscounted. The PtthDelta mutant, which has a cassette replacing that 600nt, was confirmed by PCR and by the complete absence of immunostaining against Ptth protein. However, we did not confirm the precise deletion/recombination site by DNA sequencing. As a result, PtthDelta should map to 2L:576323..576922 (as you also found out) but it is possible that the homologous recombination process made a slight mistake (of a few nucleotides).
