Dredd alleles resequencing report: Five EMS alleles of Dredd are re-sequenced to verify the positions of the molecular lesions. These alleles were originally reported by Leulier et al, EMBO Rep, 2000 Oct 16; 1(4): 353–358 (FBrf0133276). Allele DreddD44 : DreddD44 was reported to be a missence mutation at position X: 635,088, a G to C nucleotide substitution that results in a codon change from GGT (Glycine, G) to CGT(Arginine, R). Resequencing the strain reveals an additional point mutation 31 base-pairs “upstream” of the original mutation in the Dredd ORF, at the genomic position X:635 ,119. It is a T to A nucleotide change that results in ATT (Isoleucine, I) to AAT (Asparigine, N). Therefore, the DreddD44 strain in fact bears two point mutations. Allele DreddB118 : The re-sequencing result confirms the presence of the reported DreddB118 mutation, at genomic position X:635 ,067. It is a C to T nucleotide change that creates a premature stop codon ( CGA, R to TGA, STOP), implying protein truncation. Allele DreddD55 : The presence of the originally reported DreddD55 allele is also confirmed by resequencing. It is at X:634 ,862. A G to A nucleotide substitution alters the codon from TGG (Tryptophan, W) to TAG (STOP). Thus, it is also a truncated protein. Allele DreddL23 : This missence mutation is also confirmed. At genomic position X:633 ,948, there is a T to A substitution, leading to codon change from TGG (Tryptophan, W) to AGG (Arginine, R) Allele DreddF64 : In the Leulier et al report, DreddF64 is a frame-shift mutation at genomic position X:634 ,963, which affects the protein sequence at Glycine 139 in the Dredd ORF. After re-sequencing, it is revealed that the Leulier lab DreddF64 strain does not carry this mutation. It is not found in any of the sequenced Dredd allele stocks. Instead, a C to T substitution is found at genomic position X:633 ,735, which creates a premature codon that removes the last 8 amino acids of the Dredd protein (CAG, glutamine, Q, to TAG, STOP). This change affects the caspase-8 subunit p10 domain of Dredd protein and this mutant allele, behave as a genetic null and does not complement other Dredd mutants.
