The wake gene is located at 3R:22630971..22692598 (61.63 Kb) on the (+) strand. We mapped the P{UAS-Mob2.ECFP}wake32099 insertion between 3R:22666897..22666898 on the (+) strand. There is a mistake in the description of wake15185. It was originally designed two CRISPR target sites to produce wake-Gal4 line, we incorrectly described as the other one. The CRISPR Target Site 'Detailed Information' should be CRISPR Target Site PAM: ATGGAGCAGGATGCCGCAGCAGG CRISPR Target Strand Relative to Gene: minus Cutting Site: 17 bases after ATG of wake-RG Guide RNA Primers: Sense oligo 5’-CTTCGATGGAGCAGGATGCCGCAGC Antisense oligo 5’-AAACGCTGCGGCATCCTGCTCCATC Upstream Homology Arm: 1024 bp, base -1024~-1 relative to ATG of wake-RG Forward oligo 5’-TAGAAGTCACCAGCGACAGC Reverse oligo 5’-GTTGGCTGTGCGAATGCT Downstream Homology Arm: 1062 bp, base +8~+1069 relative to ATG of wake-RG Forward oligo 5’-AGGATGCCGCAGCAGGTC Reverse oligo 5’-GCATCCTGGCCACCAAATG In wake15185, bases 1-20 from the ATG ( 3R:22652428..22652447 ) have been deleted and replaced by the knock-in-cassette RMCE-Gal4-w with an attP, Gal4::VP16 , floxed white, and an inverted attP.
