The following information accompanied stocks donated to the Bloomington Drosophila Stock Center by the FlyLight Project Team and Barry Dickson, Howard Hughes Medical Institute, Janelia Research Campus. Using a strategy similar to the one used for generating the fru-GAL4 allele (FBti0168666, FBrf0191488), gene targeting by homologous recombination (Rong and Golic, FBrf0145710) was used to insert either the GAL4 DNA-binding domain or the p65 activation domain into the fru locus. Fru-specific gene targeting vectors were constructed using hemidriver sequences from pZpGAL4DBD or p65ADZp. These insertion alleles incorporate a transcription stop sequence that prevents transcription of sex-specific fru mRNAs from the P1 promoter, while preserving essential transcripts from the P2-P4 promoters. As a result, the following insertions are viable but result in male sterility when homozygous. TI{GAL4(DBD)::Zip-}fruP1.G4DBD is an insertion on the third chromosome TI{p65(AD)::Zip+}fruP1.p65AD is an insertion on the third chromosome.
