GAL80ts(int-TS19) is a temperature-sensitive version of the Saccharomyces cerevisiae GAL80 gene ( SGDID:S000004515 ), generated by inserting the temperature-sensitive TS19 intein (engineered sequence generated by mutating the VMA intein from the Saccharomyces cerevisiae VMA1 gene) into the GAL80 coding sequence. At the permissive temperature (18[o]C), the intein is functional and autonomously splices out of the coding sequence, resulting in a functional GAL80 repressor that can act as a negative regulator of the Saccharomyces cerevisiae GAL4 transcriptional activator, by binding to the GAL4 protein. At the non-permissive temperature (29[o]C), the intein is not functional and no splicing occurs, preventing the production of functional GAL80. (FBrf0179533).
