The Gal4 coding sequence was separated from the btl enhancer by a flip-out cassette containing the sequence of y (including transcriptional stop signals) flanked by FRT recombination sites to avoid constitutive expression of the transcriptional activator Gal4 in the entire tracheal system. Induction of the FLP recombinase with a mild heat shock resulted in the removal of the y cassette in individual cells, allowing for the transcription of the Gal4 gene in a small fraction of btl-Gal4 positive cells.
