10x UAS regulates expression of the artificial transcriptional activator dCas9::VPR, which is composed of a nuclease-dead form of Spyo\Cas9 fused to a highly active transcription activation sequence. The driver is tagged at the N-terminus with 3x Tag:FLAG and nuclear localization signals are present either side of the mutated Spyo\Cas9 sequence. This driver is designed to be used to overexpress a target gene of interest from its endogenous genomic locus, by combining it with sgRNAs that target sequences upstream of the transcription start site of the gene of interest.
