UASt regulatory sequences drive expression of a chimeric Hsap\TARDBP protein that consists of the N-terminal 100 amino acids (this contains the nuclear localization sequence), the linker region that is normally found between the two RNA recognition motifs (RRMs), a mutated form of RRM2 that cannot bind RNA but retains the nuclear export sequence, and 12 tandem copies of its prion-like Q/N-rich region. The protein is tagged at the N-terminal end with Tag:FLAG.
