Expression of pumScer\UAS.cSa under the control of Scer\GAL4ChAT.7.4 induces a seizure phenotype in third instar larvae subjected to an electroshock - the larvae become temporarily paralyzed with recovery time significantly longer compared to controls. Scer\GAL4eve.RRa-driven expression leads to a significant reduction of the persistent voltage-gated Na[+] current (I[[NaP]]) in aCC neurons, the transient voltage-gated Na[+] current (I[[NaT]]) is not changed compared to wild-type controls and the persistent-to-transient ratio is decreased.
Expression of pumScer\UAS.cSa under the control of Scer\GAL4ey-OK107 causes a severe defect in the mushroom body neuron axon projection. The mushroom body lobe structure is not formed and the Kenyon cell axons terminate prematurely just medial to the peduncle. No defects are observed when pumScer\UAS.cSa is expressed under the control of Scer\GAL4238Y, Scer\GAL4c739, Scer\GAL4c747 or Scer\GAL4c309.
Expression of pumScer\UAS.cSa under the control of Scer\GAL4eve.RN2O results in a reduction of both transient and persistent components of the voltage-gated Na+ current in aCC/RP2 neurons. Endogenous membrane excitability, determined by injection of constant current, is reduced.
Expression of pumScer\UAS.cSa when driven by Scer\GAL4elav-C155 prevents long term facilitation (LTF) onset at the neuromuscular junction. These larvae respond to nerve stimuli with excitatory junctional potentials (ejp's) only 20% of the time (as compared to about 80% in controls) in low Ca2+ (0.15mM) conditions. When pumScer\UAS.cSa is driven by Scer\GAL4elav-C155 no detectable behavioural defects are seen.

Expression of pumScer\UAS.cSa under the control of Scer\GAL4ChAT.7.4 or Scer\GAL4elav.PLu fully suppresses the seizure-like phenotype (temporal paralysis) observed in parabss1 mutant males subjected to brief vortexing or in mutant third instar larvae upon an electroshock and their recovery time is comparable to that of wild-type controls. Similarly, expression of pumScer\UAS.cSa driven by Scer\GAL4elav.PLu significantly reduced the seizure duration in parabss1 larvae. parabss1 mutant larvae expressing pumScer\UAS.cSa in the aCC motoneuron controlled by Scer\GAL4eve.RRa show a striking reduction in the persistent voltage-gated Na[+] current (I[[NaP]]) in the aCC but no significant change in the transient voltage-gated Na[+] current (I[[NaT]]) relative to parabss1 mutants alone, therefore the persistent-to-transient ration is markedly decreased, consistent with the presumed anticonvulsant role of pum upregulation. On occasion, multiple resurgent currents appear during the I[[NaP]] plateau in parabss1 larvae and the proportion of cells showing these are decreased upon pumScer\UAS.cSa expression.
