Expression of SRm160KK108396 alone or in combination with SRm160GL00538 (using UAS-dicer2 to enhance the RNAi efficiency) under the control of either the Scer\GAL4P2.4.Pdf or the Scer\GAL4tim.PE driver results in strong reduction in the proportion of rhythmic flies and decrease in the rhythmic power of the behavioral oscillations under free-running (constant darkness) conditions in the adult locomotor activity assay. Under driven light-dark conditions, SRm160 knockdown does not lead to any significant irregularities in the locomotor activity, the morning and evening anticipatory behavior is slightly diminished but not statistically significant. Scer\GAL4P2.4.Pdf-controlled expression of SRm160KK108396 does not lead to any structural defects to the LNv neuronal projections but it abrogates the oscillations of Pdf protein levels in the projections of s-LNv neurons and also dampens the oscillation of the per protein in these neurons.

Expression of SRm160KK108396 limited to adulthood by employing the Scer\GAL4Pdf.Switch1a driver and feeding the flies RU486 to activate the expression also results in significant decrease in the percentage of rhythmic flies.
