Molecular analysis confirms the presence of a P-element insertion just proximal to vlc. No evidence is found by polymerase chain reaction (PCR) and sequencing within 1.5 kb of the longest ap transcribed region for the ~200 bp deletion reported in FBrf0057363.
Contrary to a previous report (FBrf0057363), hemizygous ap41F flies have normal wings and halters.
Abnormal wing articulation results in flightlessness.
Weak mutant allele. Arose during a P{Δ2-3}99B mobilization of the PZ enhancer trap element to generate lines expressing Ecol\lacZ in subsets of muscles and neurons. This chromosome carries an insertion approximately 4.5kb downstream of the ap transcription unit, unrelated to the ap phenotype: 80 independent excision derivatives (60% of which were homozygous viable and 40% of which were recessive lethal) retained the ap41F phenotype.
It is suggested that ap41F is not a mutant allele of ap. First, contrary to a previous report (FBrf0057363), hemizygous ap41F flies have normal wings and halters. Second, although molecular analysis confirms the presence of a P-element insertion just proximal to vlc, polymerase chain reaction (PCR) and sequencing fails to provide evidence for the ~200 bp deletion reported in FBrf0057363 within 1.5 kb of the longest ap transcribed region.
