wing & microchaeta | distal
Overexpression of fzI.hs at 24 hours after pupariation results in a dominant gain-of-function planar cell polarity phenotype in the wing.
Heat induced expression, by waxing the wing, causes a strong tissue polarity phenotype across the wing. Application of hot wax in distal wing regions causes a dramatic hair polarity phenotype, reverse polarity is observed. Little or no effect is seen after waxing of proximal regions.
Heat shock 6 or more hours before prehair initiation results in phenotypes that resemble those of fz, dsh or pk - hsfz-early phenotype. Heat shock just before prehair initiation results in distinct phenotypes that resemble those of fy or in mutants - hsfz-late phenotype. The late phenotype can be distinguished by the dramatic difference in the number of multiple hair cells (MHCs). dsh1 but not pk1 can suppress the MHC phenotype. The early phenotype can be suppressed by reduction in dsh+ gene dosage and enhanced by an increase.
Heat shock inductions between 0 and 36 hours after pupariation resulted in abnormal wing hair polarity and hair number phenotypes similar to those of tissue polarity mutants. Rough eye and weak thoracic bristle phenotypes also result. Heat shock after 36 hours has no effect. Early heat shocks (before 30hours) result in major polarity disruptions and a few multiple hair cells. Late heat shocks (33-36hrs AP) result in numerous multiple hairs as well as abnormal polarity. An active endogenous fz gene is not required for these effects. Clonal analysis reveals a non-cell autonomy of the hair polarity and multiple hair cell phenotype.

Heat shock inductions at 37oC for 1 hour dramatically rescues the adult mutant phenotype. The best rescue is obtained when flies are raised at 29oC, though this rescue is not complete. 94% of the polarity and multiple hair cell phenotype is rescued in otherwise null mutants, but rescue is 100% for the hypomophic fz24. Leg and eye phenotypes are also rescued.
