Pan-neural expression of NetAScer\UAS.cHa under the control of Scer\GAL4sca-109-68 has no effect on apoptosis in the thoracic and abdominal segments of the ventral nerve cord.

When expression is driven by Scer\GAL4GMR.PF, retinal axons stall in the optic stalk, spread abnormally or criss-cross each other along the D/V axis. Photoreceptor axons also collapse onto each other along the A/P axis. They fail to innervate their target region properly. When expression is driven in clones in the anterior, undifferentiated portion of the eye disc or in the lamina anlage by Scer\GAL4Act5C.PP there is no effect on retinal fibers. However when clones fall in the differentiating portion of the lamina, mild hypoinnervation and irregular spacing of retinal fibers occurs.

Scer\GAL4F63-mediated expression elicits inappropriate innervation by the transverse nerve (ectopic innervation of muscles 7 and 6 and occasionally 13) but can rescue the embryonic phenotype of Ts(1Lt;YSt)B118+Ts(1Rt;YLt)T9. Scer\GAL4how-24B-mediated expression allows the axons of the transverse nerve to ectopically innervate muscles 7 and 6.

Df(1)KA9 and Df(1)RK5 embryos exhibit variable defects in the formation of commissural pathways, Scer\GAL4rho.PL induced expression of P{UAS-NETA} significantly rescued the CNS phenotype. Scer\GAL4sca-537.4 induced expression leads to a variety of CNS defects; reductions in the number of axons in commissural bundles and more extremely missing commissures.

Loss of fra function suppresses the ectopic innervation of muscles 7 and 6 by the transverse nerve caused by Scer\GAL4how-24B-mediated expression of NetAScer\UAS.cHa. Scer\GAL4how-24B-mediated coexpression of NetAScer\UAS.cHa and Sema-2aScer\UAS.cKa allows correct innervation by the transverse nerve and the RP3.

Expression of NetAScer\UAS.cHa under the control of Scer\GAL4sim.P3.7 almost completely rescues the axon scaffold defects seen in stage 16 Df(1)NetABΔ mutant embryos, although a few minor defects are still visible. Expression of NetAScer\UAS.cHa under the control of Scer\GAL4rho.NEE does not rescue the phenotypes, and commissural and longitudinal defects are still present. Expression of NetAScer\UAS.cHa in post-mitotic neurons under the control of Scer\GAL4AD.elav.T:Zzzz\ZipEERRL,T:SV40\nls2 enhances the Df(1)NetABΔ axon phenotypes, whereas the increased apoptosis seen in Df(1)NetABΔ mutant embryos from stage 14 onwards is partially rescued when NetAScer\UAS.cHa is expressed under the control of Scer\GAL4AD.elav.T:Zzzz\ZipEERRL,T:SV40\nls2, with embryos displaying an intermediate phenotype that is not statistically significant from either wild type or Df(1)NetABΔ.

Expression of NetAScer\UAS.cHa under the control of Scer\GAL4sim.P3.7 rescues the negative geotaxis defects seen in Df(1)NetABΔ mutant flies. No rescue is seen when NetAScer\UAS.cHa is expressed under the control of either Scer\GAL4rho.NEE or Scer\GAL4AD.elav.T:Zzzz\ZipEERRL,T:SV40\nls2. Expression under the control of either Scer\GAL4AD.elav.T:Zzzz\ZipEERRL,T:SV40\nls2 and Scer\GAL4sim.P3.7 (but not Scer\GAL4rho.NEE) is able to rescue the mechanical-startle induced locomotor defects.

NetAScer\UAS.cHa fails to rescue Df(1)NetABΔ when expressed under the control of any of Scer\GAL4AD.elav.T:Zzzz\ZipEERRL,T:SV40\nls2, Scer\GAL4rho.NEE, Scer\GAL4sca-109-68 or Scer\GAL4sim.P3.7.

Expression of NetAScer\UAS.cHa under the control of any of Scer\GAL4AD.elav.T:Zzzz\ZipEERRL,T:SV40\nls2, Scer\GAL4rho.NEE or Scer\GAL4sim.P3.7 has no effect on the male courtship 'mirror movement' phenotype seen in Df(1)NetABΔ mutants.

Expression of NetAScer\UAS.cHa under the control of Scer\GAL4AD.elav.T:Zzzz\ZipEERRL,T:SV40\nls2 is able to rescue the flightlessness seen in Df(1)NetABΔ mutant flies. No rescue is seen when NetAScer\UAS.cHa is expressed under the control of either Scer\GAL4rho.NEE or Scer\GAL4sca-109-68.
