    Comment:

Site of nucleotide substitution in mutant inferred by FlyBase based on reported amino acid change. Curator note: The mutation was reported to be at position 126 with respect to transcript RA, but Figure 3 makes it clear that the position is relative to transcripts RB, RC, RE, and RF, but not RA.

In contrast to control flies, ine2 mutant flies die within 10 days on hypertonic media. As in wild type, no tissue damage is observed in the anterior hindgut.

The median survival rate of ine2 mutant flies in response to lack of water is reduced compared to wild type controls. Survival rate following starvation (in the presence of sufficient water) is unchanged compared to controls.

ine2 flies maintained for 4 days on food medium containing 0M, 0.1M or 0.2M NaCl have similar viability to control wild-type flies. Flies maintained on food medium containing 0.4M NaCl show only slight viability (in contrast to wild type flies, where 90% survive). The flies also show increased sensitivity to KCl and sorbitol compared to wild-type flies.

Expression of ineRA.Scer\UAS in the hindgut under the control of Scer\GAL4Irk2.PL rescues the lethality seen when ine2 mutant flies are raised on hypertonic medium. Expressing ineRA.Scer\UAS in neurons (Scer\GAL4elav.PU) or glia (Scer\GAL4repo.PU) does not rescue the lethality.

Expression of ineRB.Scer\UAS in the hindgut under the control of Scer\GAL4Irk2.PL rescues the lethality seen when ine2 mutant flies are raised on hypertonic medium. Expressing ineRB.Scer\UAS in neurons (Scer\GAL4elav.PU) or glia (Scer\GAL4repo.PU) does not rescue the lethality.
