Description

Amino acid replacement: Q959term.

The premature stop codon falls within the third leucine zipper and is predicted to result in a 190 amino acid truncation of the protein.

Nucleotide substitution: C2875T.

cnnmfs3/Df(2R)BSC306 results in lethality with escapers.

Embryos produced by cnnmfs3/Df(2R)cnn; cnnΔEx1A females exhibit multiple nuclei and spindles scattered throughout the embryo (deep and cortical) and frequent nuclear fusions. These embryos produce normal triploid polar bodies following the completion of meiosis. In the mutant embryos, early spindles frequently have an obvious spindle pole, but the poles are lost as development proceeds and barrel-shaped anastral spindles accumulate, accompanied by nuclear fusions.

The number of adults that develop to adulthood represents only 32% of eggs laid by cnnmfs3 mutant females that also express cnnPA.Scer\UAS.P\T.T:Avic\GFP under the control of Scer\GAL4nos.UTR.T:Hsim\VP16.

The number of adults that develop to adulthood represents only 1% of eggs laid by cnnmfs3 mutant females that also express cnnS22A.PA.Scer\UAS.P\T.T:Avic\EGFP under the control of Scer\GAL4nos.UTR.T:Hsim\VP16.

The number of adults that develop to adulthood represents only 1% of eggs laid by cnnmfs3 mutant females that also express cnnT27A.PA.Scer\UAS.P\T.T:Avic\EGFP under the control of Scer\GAL4nos.UTR.T:Hsim\VP16.

The number of adults that develop to adulthood represents only 14% of eggs laid by cnnmfs3 mutant females that also express cnnS22A.T27A.PA.Scer\UAS.P\T.T:Avic\EGFP under the control of Scer\GAL4nos.UTR.T:Hsim\VP16.

Only 37% of testis somatic cyst stem cells show repositioning of the mitotic spindle during anaphase in cnnmfs3/cnnHK21 males compared to 78% of cases in heterozygous controls.

The failure of embryogenesis in mutant animals is associated with the appearance of multiple metaphase spindles fused at a common pole.

The number of germ cells associated with the hub is increased 20-30% in homozygous males, from an average of 8.94 GSCs per hub in wild type to 10.69 GSCs per hub in cnnHK21/cnnmfs3 males. In some cases in cnnHK21/cnnmfs3 testes, a GSC divides and the two daughter cells remain in contact with the hub (this is not observed in the wild type).

Homozygous females produce embryos that arrest prior to cellularisation. Embryos derived from cnnmfs3/Df(2R)cnn females have severe defects in nuclear division and distribution. The embryos never achieve cellularisation.

Cytokinesis and karyokinesis during meiotic divisions are disrupted in males. The percentage of aberrant spermatids varies from testis to testis (20-50%), and the cytokinetic defects are more severe than karyokinetic defects.

cnnmfs3 is partially rescued by cnnhs.PL

Comments

Expression of cnnPA.Scer\UAS.P\T.T:Avic\GFP under the control of Scer\GAL4nos.UTR.T:Hsim\VP16 rescues embryogenesis in cnnmfs3 animals; approximately 92% of eggs produced by cnnmfs3 females expressing cnnPA.Scer\UAS.P\T.T:Avic\GFP under the control of Scer\GAL4nos.UTR.T:Hsim\VP16 hatch.

Male and female sterility is rescued by cnn+t16. Male sterility is rescued by basal levels of expression (at 25oC) of cnnhs.PL.
