In a background in which vgScer\UAS.cZa is expressed under the control of Scer\GAL4salm-EPv, Scer\GAL4ap-md544 or Scer\GAL4nub.PK, cell clones fail to grow preferentially along the proximal-distal axis (as they do in a wild type background) and the orientation of cell divisions is perturbed.

Expression of vgScer\UAS.cZa in the posterior compartment of the wing imaginal disc under the control of Scer\GAL4en-e16E results in a 15% reduction in the number of cells in G1 compared to the anterior compartment of the disc, and increased cell proliferation. There is no difference in average cell size compared to control cells.

Expression of vgScer\UAS.cZa under the control of Scer\GAL4Dll-md23 results in the autonomous transformation of distal territories of the appendages (proboscis, first and second pairs of legs and genitalia) into tissues with characteristics of the wing blade. In the first and second pairs of legs, these transformations produce trichomes, anteroposterior and dorsoventral wing margin chaetae, wing veins and campaniform sensilla. The transformed legs have large ventral wing territories and small dorsal territories encircled by wing margin. In the third pair of legs, transformation to haltere tissues is seen. Expression of vgScer\UAS.cZa under the control of Scer\GAL4Dll-md23 results in the transformation of antenna with anterior and posterior wing margin elements. Expression of vgScer\UAS.cZa under the control of Scer\GAL4vg.PM does not result in cuticular transformation of the eye. Expression of vgScer\UAS.cZa under the control of Scer\GAL4Ubx.PdC in clones in the wing results in tubular, perpendicular outgrowths to the wing surface. The outgrowths contain both clone cells and surrounding wild-type cells. The clones are frequently located at the tip of the outgrowth but they can also grow along the lateral zones. In those clones that appear in central parts of the wing, all the cells of the outgrowth and the clone always show a differentiation corresponding to wing blade trichomes. Clones near the wing margin may differentiate marginal chaetae. Clones in the notum expressing vgScer\UAS.cZa under the control of Scer\GAL4Ubx.PdC contain wing blade trichomes and notum chaetae in a "salt and pepper" distribution. Clones in the leg expressing vgScer\UAS.cZa under the control of Scer\GAL4Ubx.PdC have "mixed" cuticular differentiation patterns, with leg chaetae being mixed in a "salt and pepper" distribution with wing blade trichomes.

Expression of vgScer\UAS.cZa under the control of Scer\GAL4ptc-559.1 causes deformation of the wing disc.

Animals expressing vgScer\UAS.cZa under the control of Scer\GAL4ptc-559.1 usually die at the early pupal stage. Animals that die at the late pupal stage show extensive wing-like outgrowths associated with a disorganisation and reduction of the wings. The formation of ectopic wing-like outgrowths caused by expression of vgScer\UAS.cZa under the control of Scer\GAL4ptc-559.1 is more strongly reduced in a vg1, vg83b27 or vgnull homozygous background than in the corresponding heterozygous vg1, vg83b27 or vgnull background.

Co-expression of vgScer\UAS.cZa in flies expressing dapScer\UAS.cLa at the dorsal/ventral wing boundary under the control of Scer\GAL4vg.int2.1 in a vgnull/+ mutant background rescues wing morphology, and mitotic activity of cells is restored in this region.

The transformation of the legs and antenna seen in flies expressing vgScer\UAS.cZa under the control of Scer\GAL4Dll-md23 is strongly reduced by coexpression of wg-C.Scer\UAS. Co-expression of vgScer\UAS.cZa and wgScer\UAS.cLa under the control of Scer\GAL4vg.PM causes the transformation of the eye into wing blade, resulting in outgrowths with histotypical characteristics typical of wing blade. Co-expression of vgScer\UAS.cZa and wgScer\UAS.cLa under the control of Scer\GAL4Ubx.PdC in clones in the wing imaginal disc leads to the formation of tubular and perpendicular outgrowths from the wing blade. The size of the outgrowths is larger than in clones expressing vgScer\UAS.cZa under the control of Scer\GAL4Ubx.PdC and is dependent on the distance from the wing margin. All the cells of the wg-vg expressing clones are differentiated into wing margin sensory elements, whereas non-autonomous territories of the outgrowth are differentiated into wing blade trichomes. Co-expression of vgScer\UAS.cZa and wgScer\UAS.cLa under the control of Scer\GAL4Ubx.PdC in clones in the wing hinge or notum cause transformation phenotypes; the cells autonomously differentiate into wing margin and non-autonomously differentiate into wing blade trichomes. Co-expression of vgScer\UAS.cZa and wgScer\UAS.cLa under the control of Scer\GAL4Ubx.PdC in clones in the leg results in a "mixed" phenotype with leg chaetae mixed with wing blade trichomes in a "salt and pepper" distribution. Co-expression of vgScer\UAS.cZa and wgScer\UAS.cLa under the control of Scer\GAL4Ubx.PdC in clones in the eye results in the cells autonomously differentiating into sensory elements of the wing margin.

Marker expression data suggests that the presence of vgScer\UAS.cZa transforms wgScer\UAS.cGa; Scer\GAL4αTub84B.PL somatic clone cells in the peripodial epithelium from wing hinge like to wing pouch like.

Females expressing vgScer\UAS.cZa under the control of Scer\GAL4ptc-559.1 in a sdETX4/+ background die in the late pupal stage. They show ectopic wing-like outgrowths at the posterior of the head capsule, with strong deformation of the eye. Males expressing vgScer\UAS.cZa under the control of Scer\GAL4ptc-559.1 in a hemizygous sdETX4 background do not show such extensive wing-like outgrowths and show only slight deformation of the eye. These males can emerge from the pupae, although they die within a few hours.

Expression of vgScer\UAS.cZa under the control of Scer\GAL4ptc-559.1 in vgnull mutant discs increases the number of cells in S-phase and results in overgrowth of the disc along the anterior/posterior axis. An increase in cells in M-phase is observed.

Scer\GAL4ap-md544; vgScer\UAS.cZa; partially but significantly rescues the adult muscle phenotype vgnull/vgnull flies.

The wing defects seen in vg83b27 flies are completely rescued by expression of vgScer\UAS.cZa under the control of Scer\GAL4vg.PU.
