    stnA1 stnB1/Y ("stn8P1") males do not survive to adulthood. stnA1 stnB1 third instar larvae show a reduced mean excitatory junction potential (EJP) amplitude at the neuromuscular junction compared to wild-type larvae. stnA1 stnB1 ("stn8P1") larvae show dramatic alterations in presynaptic architecture at the neuromuscular junction; small bud-like "satellite" boutons are seen proliferating from normal boutons on the presynaptic motor terminals. The lethality of stnA1 stnB1/Y ("stn8P1") animals is not rescued by expression of sytScer\UAS.cLa under the control of either Scer\GAL4elav-C155 or Scer\GAL4shi.PS.

    Adult stnA1 stnB1 males survive at a frequency of approximately 1% relative to their control siblings. mejp frequency at the stnA1 stnB1 larval neuromuscular junction is nearly identical to that of controls. EJPs evoked by stimulation of the motor nerve are reduced to approximately 10% of wild type. This decreased EJP amplitude derives from a severe reduction in quantal content (4.7 +/- 0.7 compared to the wild-type 133.9 +/- 16.1). Average mejp amplitude and distribution of mejp amplitudes are not increased. The boutons of the stnA1 stnB1 larval neuromuscular junction show a 2.6-fold decrease in synaptic vesicle density compared to controls. The vesicles are on average 44.8 +/- 1.1 nm in diameter (compared to 34.4 +/- 0.6 nm for controls), corresponding to an approximately 2-fold increase in mean vesicle volume and in vesicle size variability.
