UAS sequences drive expression of an intracellular fragment of N, which encompasses the cdc10/ANK repeats. The first two amino acids of the cact protein followed by a 14 amino acid Tag:MYC epitope which is then fused to amino acids 1895--2156 of N.

Somatic clones expressing Ncdc10.UAS.Tag:MYC under the control of Scer\GAL4Tub.PU (in combination with a Gal80[ts] transgene to restrict expression to the adult stage) contain a smaller overall number of cells and intestinal stem cells in adult flies compared to controls.

Adulthood-generated midgut clones expressing Ncdc10.UAS.Tag:MYC under the control of Scer\GAL4Tub.PU have significantly fewer cells, namely intestinal stem cells, than control clones.

The number of intestinal stem cells (ISCs) in the posterior midgut of flies expressing Ncdc10.Scer\UAS.T:Hsap\MYC under the control of Scer\GAL4esg-NP5130 15 days after hatching is decreased compared with wild type.

Expression of Ncdc10.Scer\UAS.T:Hsap\MYC driven by Scer\GAL4ptc-559.1 prevents the development of the scutellar bristles.

Adulthood-generated spen9C95 midgut clones show a severe increase in the number of cells, namely of Intestinal stem cells (Dl-positive), as compared to control clones; both are suppressed if the clones express Ncdc10.UAS.Tag:MYC under the control of Scer\GAL4Tub.PU. When Ncdc10.UAS.Tag:MYC expression is induced only 10 days after clone induction, the clones become composed of enterocytes.

Flies co-expressing SaraGD8765 and Ncdc10.Scer\UAS.T:Hsap\MYC under the control of Scer\GAL4esg-NP5130 have a reduced number of intestinal stem cell compared to controls, as is seen when Ncdc10.Scer\UAS.T:Hsap\MYC is expressed alone.

Expression of Ncdc10.Scer\UAS.T:Hsap\MYC under the control of Scer\GAL4tub.PU suppresses the formation of supernumerary intestinal stem cell-like cells which are produced in homozygous GmdH78 clones in the adult intestine.

Expression of Ncdc10.Scer\UAS.T:Hsap\MYC driven by Scer\GAL4ptc-559.1 has no effect on the Su(H)8/Su(H)2 proneural cluster phenotype in the wing disc.
