Description

The stau cDNA, which has had the amino acids that divide double stranded RNA Binding Domain 2 (dsRBD2) into two halves replaced by the corresponding 8 amino acid loop of dsRBD3, is driven by the αTub67C promoter and tagged at the amino terminus with Tag:MYC.

Embryos that express both miraΔ103.αTub67C.T:Avic\GFP-m6 and stauΔloop2.αTub67C.T:Hsap\MYC in a stau2 background lack a posterior abdomen and form an anterior abdomen with complete reverse polarity.

Comments

Addition of stauΔloop2.αTub67C.T:Hsap\MYC to stau2 mothers gives little or no rescue of the stau posterior phenotype, none of the progeny of these flies hatch into larvae, and almost all develop less than one abdominal segment. However, the head skeleton defects seen in stau2 flies are rescued: 2/3 of embryos have wild-type heads (compared to none in stau2 alone) and the rest have much milder head defects.
