UAS regulatory sequences drive expression of InR that carries an K418P amino acid replacement (numbering using GenBank submission gb|AAC47458). This change corresponds to K414P in the current release of the Drosophila genome (release 6). The change is meant to mimic the R113P change in Longo et al., 1993 (PMID:8419945) (R113P is called R86P in the paper). Expression of InRR418P.Scer\UAS under the control of Scer\GAL4E(spl)m6-BFM.PF leads to significantly increased numbers of abdominal lateral and dorsal, but not ventral, adult muscle precursor cells in third instar larvae, as compared to controls; this expression does not affect the number of mitotic adult muscle precursor cells at embryonic stages, as compared to controls.
Fasted third instar larvae expressing InRR418P.Scer\UAS under the control of Scer\GAL4NPFR1.6.6 display an attenuated feeding response to solid food. These third instar larvae do not exhibit significant changes in the intake rate of the richer liquid food, relative to controls.
