Mutants hold their wings erect (indicative of mitochondrial dysfunction and muscle degeneration) when maintained either under normal conditions or hyperoxia. Mutants also show a marked reduction in lifespan relative to controls in response to hyperoxia.

Under hyperoxia, the mean survival time of SdhBEY12081 flies is reduced to 10% of that of normal flies.

Young flight muscles in SdhBEY12081 mutants do not display major defects in the flight muscle mitochondria, but after 15 hours of hyperoxia exhibit severe mitochondrial pathology. The majority of mitochondria are severely damaged and their cristae in disarray, whereas the myofibrils appear unaffected.

Under normal conditions SdhBEY12081 flies display a 66% decrease in mean survival time and a 17% decrease in maximum survival time.

At 2 days of age, SdhBEY12081 mutant flies appear normal in climbing assays, but this rapidly declines with age. At 8 days, mutant flies display a 54% decrease, and at 12 days they exhibit a 77% decrease in climbing score, whereas normal flies show no decline in that time.

Complex I-specific (pyruvate/malate-dependent/rotenone-sensitive) respiration rate in normal in SdhBEY12081 mutants, as is the complex IV-specific (ascorbate/TMPD-dependent/KCN-sensitive) respiration rate. However, there is a 56% decrease in the complex II-specific (succinate-dependent/antimycin-A-sensitive) respiration rate. Complex II-mediated electron transer is reduced by 40% in SdhBEY12081 mutants. Mitochondria from SdhBEY12081 mutants exhibit a 32% increase in hydrogen peroxide production, as compared to wild-type controls.
