Detailed Description
Statement

Mutant flies are less sensitive than wild-type flies to acute heat stress, with the time to total paralysis being significantly longer in the mutant flies than in wild-type controls.

At 38[o]C, motor neurons of mutant larvae are unable to increase their firing rate in response to heat stress, in contrast to wild-type controls. The mutant larvae do not show abnormal seizure-like locomotion at this temperature, in contrast to that seen in wild-type controls.

Homozygous and ppk29f06838/Df(2R)BSC136 males show significantly reduced levels of courtship (both courtship index and the fraction of males initiating courtship in the presence of decapitated females) compared to wild-type males.

Homozygous males do not show significant levels of courtship towards other males.

Expression of ppk25Scer\UAS.cSa under the control of either Scer\GAL4ppk29.3.020 or Scer\GAL4ppk25.PS does not rescue the reduced courtship index of ppk29f06838/ppk29f04205 males.

Comments

The increased time to total paralysis in response to heat stress which is seen in ppk29f06838 flies is fully rescued by expression of ppk293'UTR.Scer\UAS under the control of Scer\GAL4elav-C155.

The increased time to total paralysis in response to heat stress which is seen in ppk29f06838 flies is partially rescued by expression of either ppk29Δ3'UTR.Scer\UAS or Disc\RFPmCherry.ppk29.3'UTR under the control of Scer\GAL4elav-C155.

Expression of ppk29Scer\UAS.cLa under the control of either Scer\GAL4ppk29.3.020 or Scer\GAL4ppk25.PS rescues the reduced courtship index of ppk29f06838/ppk29f04205 males. Partial rescue is seen in the absence of a Scer\GAL4 driver.

Expression of ppk29Scer\UAS.alt under the control of either Scer\GAL4ppk29.3.020 or Scer\GAL4ppk25.PS does not rescue the reduced courtship index of ppk29f06838/ppk29f04205 males.
