A 3020bp fragment from upstream of the ppk29 translation start codon is fused upstream of sequence encoding a GAL4 driver. This fragment contains 700bp of intergenic sequence between ppk29 and the neighbouring gene eEF5 and 2.3kb of ppk29 5' UTR sequence (including two large introns, non-coding exons 1 and 2 and a non-coding segment of exon 3).
ScerGAL4ppk29.3.020 drives expression in one or two of four gustatory receptor neurons of many of the taste bristles in the male legs. All of the prothoracic gustatory receptor neurons labelled by ScerGAL4ppk29.3.020 co-express EcollexAfru.P1. For the pairs of prothoracic neurons labelled by EcollexAfru.P1, expression of ScerGAL4ppk29.3.020 can be detected in both, or only one of the neurons. Expression of ScerGAL4ppk29.3.020 and ScerGAL4ppk25.PS is very similar: all but 11 ScerGAL4ppk29.3.020 positive neurons, are also labelled by ScerGAL4ppk25.PS. In most cases, these 11 ScerGAL4ppk29.3.020-expressing neurons are paired within a single taste hair with a neuron that expresses both ScerGAL4ppk25.PS and ScerGAL4ppk29.3.020.
Gustatory receptor neurons in the wing margin and labellum also express ScerGAL4ppk29.3.020. The leg and wing neurons project to each of the thoracic ganglia. The prothoracic neurons that are labelled by ScerGAL4ppk29.3.020 send sexually dimorphic projections to the first thoracic ganglia: projections from males cross the midline more often than female ones. Gustatory receptor neurons in the labellum project to a discrete area in the subesophageal ganglion.
