The PBac{RB}orb2e01925 insertion maps within both orb2 and CG43783 (the genes share some exons at the 5' end, including some coding sequence).
male sterile (with Df(3L)ED4416)
Fertility of homozygous males is substantially impaired.
11% of orb2e01925 embryos exhibit RP2 neuron specification defects. In approximately 70% of cases the RP2 neurons are duplicated and two Eve-positive cell are seen per hemisegment. However, hemisegments are also seen that completely lack Eve-positive cells. Abnormalities in asymmetric division are also observed in the P2 to EPC and P15 to DA1 lineages. Unlike in wild type, the mitotic spindle orientation of dividing orb2e01925 neuroblasts is often randomized with respect to the apical-basal axis.
The orb2e01925 mutant takes longer than wild type to return to a standing position following vortexing and fewer flies climb the vial following knockdown. The time taken for flies to recover from heat shock is similar to controls.
Precise excision of the transposon rescues the lethality seen in orb2e01925.
