intestinal stem cell & adult midgut
The half life of follicle stem cells in Usp36f01742 homozygous clones significantly reduced compared to wild-type. But mutant follicle cells develop normally.
In Usp36f01742/Usp3602331 adults, intestinal stem cells are present at near normal numbers at eclosion but are rapidly lost so that few are left by 7 days post eclosion. This phenotype is partially suppressed by the addition of 8mM DAPT (a gamma-secretase inhibitor) to the food. Unlike wild-type adults, mutants treated with DAPT do not form tumors.
The half life of female germline stem cells in Usp36f01742 somatic clones is sharply reduced compared to wild-type. Marker expression studies indicate that germ cells in the female germline stem cell begin to differentiate prematurely.
Usp36f01742 dominantly suppresses position effect variegation in the gastric secae due to Tp(3;Y)BL2.
Usp36f01742 ; bamΔ86 double homozygotes have increased numbers of female germline stem cells compared to wild-type, as bamΔ86 homozygotes do.
The midgut of 7 day old Usp36f01742 homozygous adults has greatly reduced numbers of intestinal stem cells.
