Detailed Description

The number of nuclear bundles and individualization complexes is significantly decreased in the testis of ctpexc39/Y mutants compared to controls. The F-actin is decreased in the investment cones during ctpexc39 sperm individualization compared to wild-type controls.

Weak allele. ctpexc6/ctpexc39 and ctpexc39/Df(1)bi-DL1 transheterozygotes exhibit thin bristles and erect wings. The seminal vesicles of ctpexc39 hemizygous males contain lower numbers of motile sperm than wild type. In the testes of 2 day-old ctpexc39 adult males, spermatids are abnormally knotted in the middle and coiled at their distal ends inside the cysts. The average length of cysts is shortened in ctpexc39 testes, as the peak of the elongation cone (EC) distribution profile is shifted to the basal end. The total number of ECs per testis is reduced in ctpexc39 mutants that carry one copy of Df(3L)GN24; some of these ECs are deformed and the average cyst length is shorter than in ctpexc39 single mutants. In ctpexc39/Df(3L)fz-GF3b mutants, there are almost no detectable ECs.

ctpexc39; sw1 mutants are viable at 25oC.

The ctpexc39/Y nuclear bundles and individualization complex phenotype is dominantly enhanced by Gl1. The phenotype is also dominantly enhanced by Df(3L)GN24, which uncovers the Dhc64C gene. In contrast, the Dhc64C4-19 allele does not affect this phenotype.

The sterility of ctpexc39 males is further reduced when flies carry one copy of Gl1 or Df(3L)fz-GF3b. The total number of elongation cones (ECs) per testis is reduced in ctpexc39 mutants that carry one copy of Gl1; some of these ECs are deformed and the average cyst length is shorter than in ctpexc39 single mutants. The same phenotype occurs when ctpexc39 flies carry one copy of Df(3L)GN24. In ctpexc39; Df(3L)fz-GF3b/+ mutants, there are almost no detectable ECs.
