Silencing of Tdc and dsx-positive neurons by expressing Ctet\TeTxLCScer\UAS.cSa under the control of Scer\GAL4Tdc2.PC (in which the Scer\FRT cassette has been excised by Scer\FLP1dsx) at the permissive temperature results in disrupted post-mating practices in mated females. These females are more receptive than wild type females, as they remate at a significantly higher frequency and show decreased levels of rejection behaviors (ovipositor extrusion). In addition, they elicit more male courtship and show decreased levels of egg laying.

Silencing neurons that express Ctet\TeTxLCScer\UAS.cSa under the control of Scer\GAL44-57, and in which the Scer\FRT cassette has been excised using Scer\FLP1fru, significantly delays wing extension behavior under infrared light compared to controls. Under ambient light, Ctet\TeTxLCScer\UAS.cSa expression in these neurons does not significantly alter courtship latency. The quality of courtship (the courtship index) is significantly reduced in both ambient and infrared light. Most of these flies fail to exhibit any copulatory behaviors under infrared light. The majority of these males perform wing extension/song behaviors but make no detectable attempts at copulation. No difference from controls is seen under ambient light.

Blocking synaptic transmission in neurons expressing both ppk29 and fru (through expression of Ctet\TeTxLCScer\UAS.cSa under the control of Scer\GAL4ppk29.3.612 and in which the Scer\FRT stop cassette has been removed by Scer\FLP1fru) impairs the inhibition of female contact-induced male-male aggression seen in wild type males that have previously been exposed to females.

Blocking synaptic transmission in neurons expressing both 5-HT1B and fru (through expression of Ctet\TeTxLCScer\UAS.cSa under the control of Scer\GAL45-HT1B.PY and in which the Scer\FRT stop cassette has been removed by Scer\FLP1fru) impairs the inhibition of female contact-induced male-male aggression seen in wild type males that have previously been exposed to females. Suppressing expression in the GABA+ neurons using Scer\GAL80Gad1 blocks this disinhibition.

Blocking synaptic transmission in GABA+ neurons expressing fru (through expression of Ctet\TeTxLCScer\UAS.cSa under the control of Scer\GAL4Gad1.PS and in which the Scer\FRT stop cassette has been removed by Scer\FLP1fru) impairs the inhibition of female contact-induced male-male aggression seen in wild type males that have previously been exposed to females.

Blocking synaptic transmission in neurons expressing both Rdl and fru (through expression of Ctet\TeTxLCScer\UAS.cSa under the control of Scer\GAL4Rdl.PS and in which the Scer\FRT stop cassette has been removed by Scer\FLP1fru) results in lethality or severe courtship issues.

Males expressing Ctet\TeTxLCScer\UAS.cSa under the control of Scer\GAL4ple.PF in the subset of neurons that also express Scer\FLP1383 (the Scer\FLP1 allele generates Ctet\TeTxLCScer\UAS.cSa from its progenitor Ctet\TeTxLCScer\FRT.stop.Scer\UAS, resulting in Ctet\TeTxLC expression) show very low levels of locomotion compared to controls.

Males expressing Ctet\TeTxLCScer\UAS.cSa under the control of Scer\GAL4ple.PF in the subset of neurons that also express Scer\FLP1346 show slightly locomotion defects.

Males expressing Ctet\TeTxLCScer\UAS.cSa under the control of Scer\GAL4ple.PF in the subset of neurons that also express either Scer\FLP1243 or Scer\FLP1447 show normal locomotion. These males show a change in aggressive behaviour compared to wild-type males, with a significant increase in the number of lunges, without changing the latency to the first lunge or the time to establish dominance.

Males carrying shits.Scer\UAS clones induced using Scer\FLP1fru and expressed under the control of either Scer\GAL4ple.PF, Scer\GAL4Tab2-201Y, Scer\GAL41471, Scer\GAL4Or67d-GAL4-1, Scer\GAL43-8, Scer\GAL48-194, Scer\GAL4NP0368, Scer\GAL410-16 or Scer\GAL4bl-NP3591 show significantly reduced courtship learning compared to controls.

Males carrying shits.Scer\UAS clones induced using Scer\FLP1fru and expressed under the control of either Scer\GAL4Cp1-NP7036 do not show defects in courtship learning.

Expression of Ctet\TeTxLCScer\UAS.cSa in the intersection of the Scer\GAL4dsx.KI and Scer\FLP1250 expression patterns (Ctet\TeTxLCScer\UAS.cSa generated from Ctet\TeTxLCScer\FRT.stop.Scer\UAS using the Scer\FLP1250 line) suppresses the normal postmating behavioural responses in mated females: sexual receptivity remains high, ovipositor extrusion remains low and the oviposition rates are lower compared to control mated females. In addition, these mutant mated females elicit higher levels of courtship from naive males compared to control mated females.

Males containing Ctet\TeTxLCScer\UAS.cSa; Scer\GAL4fru-GAL4/+ somatic clones induced using Scer\FLP1ey.PN paired with wild-type virgin females have low courtship index scores compared to Scer\GAL4fru-GAL4/+; Scer\FLP1ey.PN controls.
