Description

UAS regulatory sequences drive expression of an inverted repeat of a 510bp cDNA fragment encoding amino acids 68-238 of Irc. The repeated copies of this 510bp fragment are separated by an intronic spacer.

Flies expressing IrcdsRNA.Scer\UAS under the control of Scer\GAL4da.G32 show reduced survival compared to control flies after natural infection with either live or heat killed "Ecc-15" (P.carotovorum.carotovorum).

In the presence of hydrogen peroxide, the survival rates of flies expressing IrcdsRNA.Scer\UAS under the control of Scer\GAL4da.G32 are significantly lower than those of control flies. Animals expressing IrcdsRNA.Scer\UAS under the control of Scer\GAL4da.G32 show reduced survival after natural infection (feeding with contaminated food) compared to control flies. This effect has been seen with each of the following microorganisms; "Ecc-15" (P.carotovorum.carotovorum), "Ecc-15-evf-/-, E.coli, S.cerevisiae, M.luteus and S.typhimurium. Animals expressing IrcdsRNA.Scer\UAS under the control of Scer\GAL4cad-md509 show reduced survival after natural infection with "Ecc-15" (P.carotovorum.carotovorum).

Flies expressing IrcdsRNA.Scer\UAS under the control of Scer\GAL4cad-em459 show a reduced survival rate compared to control flies when naturally infected with "Ecc15" (P.carotovorum.carotovorum).

Expression of IrcdsRNA.Scer\UAS under the control of Scer\GAL4da.G32 partially suppresses the wound-induced mortality seen in Hayan1 mutants. The mortality is also rescued when IrcdsRNA.Scer\UAS is expressed in neurons under the control of Scer\GAL4Cha.7.4, but not when it is expressed in hemocytes under the control of Scer\GAL4He.PZ.

The reduced survival rate after natural infection with "Ecc-15" (P.carotovorum.carotovorum) that is seen in flies expressing IrcdsRNA.Scer\UAS under the control of Scer\GAL4da.G32 is partially rescued by pucE69/+. The reduced survival rate after natural infection with "Ecc-15" (P.carotovorum.carotovorum) that is seen in flies expressing IrcdsRNA.Scer\UAS under the control of Scer\GAL4da.G32 is partially rescued by co-expression of either CatT:mito1 or CatScer\UAS.cMa. The reduced survival rate after natural infection with "Ecc-15" (P.carotovorum.carotovorum) that is seen in flies expressing IrcdsRNA.Scer\UAS under the control of Scer\GAL4da.G32 is slightly rescued by co-expression of Jafrac1Scer\UAS.cHa.

Flies co-expressing IrcdsRNA.Scer\UAS and DuoxdsRNA.Scer\UAS under the control of Scer\GAL4cad-em459 show a normal survival rate when naturally infected with "Ecc15" (P.carotovorum.carotovorum).
