Mutant sperm also show excessive folding into tangles in the PSR. At later time points (1-4 hours after mating), the phenotype is more severe with mostly tangled flagella in the PSR lumen, and the number of mutant sperm that entering the distal seminal receptacle reduced by 100-fold.
In contrast to wild type, the number of lobolobo/loboMB00722 sperm that enter the distal seminal receptacle (DSR) vary greatly on an individual basis and the average number of sperm stored per female per mating is only 4. Only 3.9 F1 progeny are produced per female from matings to these mutant males.
Sperm from lobolobo/loboMB00722 males show head-leading orientation when they exit the proximal seminal receptacle, as wild type.
Pkd2ko67/Pkd2MB06703, lobolobo/loboMB00722 has male semi-sterile phenotype
Sperm from lobolobo/loboMB00722; Pkd2ko67 / Pkd2MB06703 double mutants are motile, are produced in large numbers and can enter the uterus as normal. However, in contrast to wild type, the number of sperm that enter the distal seminal receptacle (DSR) vary greatly on an individual basis and the average number of sperm stored per female per mating is only 2.4. Only 2.9 F1 progeny are produced per female from matings to these double-mutant males.
Stored sperm from lobolobo/loboMB00722; Pkd2ko67 / Pkd2MB06703 double mutant males can exit the distal seminal receptacle and fertilize the egg.
Excision of the Mi element reverts the mutant phenotype.
