Excision of P{EPgy2}pds5EY06473 using Δ2-3 results in a deleted sequence 3' to the insertion site removing coding sequences through to exon 5. The deletion starts 67 nucleotides upstream of the wild-type start site predicted by EST analysis.

Homozygous pds5e6 and heterozygous pds5e3/pds5e6 mutants are lethal in late third instar to early pupal stages of development. Late third instar larvae of both mutants display small or missing imaginal discs, and the larval brains are approximately half the volume of wild-type, consistent with a mitotic defect.
The vast majority of metaphases in pds5e6 are abnormal. Nearly all display aneuploidy (approximately 87.5%), and most display precocious sister chromatid separation (approximately 93.8%).
The salivary glands of homozygous pds5e6 mutants are reduced. In size-matched third instars, the pds5e6 mutant chromosomes are thinner than wild-type.
The pds5e6 mutant reduces the binding of cohesin to salivary gland chromosomes.
