D344N

Comment:

Site of nucleotide substitution in mutant inferred by FlyBase based on reported amino acid change. Nature of mutation inferred by FlyBase curator. Residue 344 is Glu (GAG) rather than Asp (GAT/C). The G to A mutation that would have given rise to the reported D344N mutation in the published strain would lead to an E344K mutation in the sequenced strain.

Detailed Description

Metaphase to anaphase transition is delayed in aurA17961/aurA14641 transheterozygous larval brain neuroblasts.

Mutants show a massive increase in larval brain neuroblasts, developing more than 1000 central brain neuroblasts at stages where wild-type larvae have only 95-100 neuroblasts.

Homozygous larvae have supernumerary neuroblasts. The mitotic spindle is often misoriented relative to the mira protein crescent in the mutant neuroblasts, in contrast to wild-type where the spindle is always aligned perpendicular to the mira protein crescent.

The increased neuroblast number and mitotic index seen in aurA8839/aurA17961 transheterozygote larvae is not enhanced by combination with BubR1KEN.T:Disc\RFP-mRFP,BubR1k06109 (the mitotic index in the double mutants is even slightly decreased), whereas the rate of ploidy defects is increased compared to either of the single gene mutants.
