Gr28bMB03888 mutant flies do not show significant changes in the expected avoidance response to food laced with acetic acid (5%), as compared to controls.

Gr28bMB03888 mutant larvae have slightly reduced rolling (escape) behavior in response to noxious temperature increases, compared to controls.

Gr28bMB03888 mutants show the expected S-b sensilla responsiveness to the bitter chemicals LOB, BER, DEN.

Gr28bMB03888 mutant adults do not have any changes in food intake under fed or starved conditions, compared to controls.

Gr28bMB03888 homozygous mutant adults do not have any substantial alterations in the responses of fast-cool or slow-cool projection neurons (PNs) to temperature changes, compared to controls; warm PNs have non-significantly reduced responses to slow, but not fast, warming, compared to controls and their responses to small warming steps are significantly reduced; unlike controls, the responses of warm PNs in Gr28bMB03888/Gr28bMB03888 mutants to fast warming or small warming steps are abolished when inhibition is blocked using picrotoxin and CGP54626.

In the absence of drugs, warm-cool PNs in Gr28bMB03888/Gr28bMB03888 mutant adults do not have any substantial alterations in responses to cooling or warming, including to small temperature steps, compared to controls; unlike in controls, warm-cool PNs in mutants lose their responses to warming, but not cooling, when inhibition is blocked using picrotoxin and CGP54626.

Gr28bMB03888/Df(2L)Exel7031 mutant flies show complete loss of the "rapid negative thermotaxis" (response to a steep temperature gradient after 1 min) seen in wild type flies. When flies are exposed to a shallow gradient for 30 minutes the flies behave similarly to wild type.

Gr28bMB03888/Df(2L)Exel7031 mutants exhibit a significant reduction in the light-induced response of class IV dendritic arborization neurons. However, the basal firing rate is not affected.

Expression of TrpA1Scer\UAS.(B).cKa in the arista under the control of Scer\GAL4Gr28b.d.PT significantly rescues the "rapid negative thermotaxis" defects seen in Gr28bMB03888 mutant flies.

Expression of TrpA1Scer\UAS.(A).cKa in the arista under the control of Scer\GAL4Gr28b.d.PT does not rescue the "rapid negative thermotaxis" defects seen in Gr28bMB03888 mutant flies.

Transheterozygotes carrying one mutant allele of TrpA1unspecified and one copy of Gr28bMB03888 exhibit a reduced light response, suggesting that Gr28b and TrpA1 function in the same phototransduction pathway.

    Comments

Expression of Gr28bScer\UAS.D under the control of either Scer\GAL4Gr28b.d.PT or Scer\GAL4hot-cell rescues the "rapid negative thermotaxis" defects seen in Gr28bMB03888 mutant flies.

Expression of Gr28b+t14.5 rescues the "rapid negative thermotaxis" defects seen in Gr28bMB03888 mutant flies.
