At a permissive temperature (18[o]C) expression of Avic\GFPdsRNA.R.Scer\UAS under the control of Scer\GAL4Cg.PA and Scer\GAL80ts.αTub84B knocks down expression of vkgG454. These flies develop into normal adults, although pupal lethality results when the larvae are transferred to restrictive temperature after completion of embryogenesis. Dissection of these larvae 96 hours after the temperature shift reveals highly aberrant shapes in several organs. Imaginal discs, consisting of highly columnar epithelial cells, appear flattened while the ventral nerve cord portion of the central nervous system is strikingly elongated, reminiscent of embryonic defects in ventral nerve cord condensation. Additionally, the ducts and imaginal rings of the salivary glands are dilated, resulting in an expanded lumen.

Adults expressing Cg25CGD12784 under the control of Scer\GAL4elav.PLu (in the presence of Dcr-2Scer\UAS.cDa to increase the efficiency of RNAi) do not show a significant defect in avoidance of noxious temperature (46[o]C) compared to control flies.

Depending on the insertion line used, expression under the control of Scer\GAL4Mef2.PR can result in early larval lethality or late larval lethality.

Expression under the control of Scer\GAL4Mef2.PR results in grossly normal larval body wall muscles.

Expression under the control of Scer\GAL4Mef2.PR results in wild-type sarcomeres in the larval body wall muscles.
