Flies expressing Gem3GD17221 (co-expressed with Dcr-2Scer\UAS.cDa) under the control of Scer\GAL4Mef2.PR show an age-dependent reduction in climbing ability compared to controls. The flies exhibit age-dependent flightlessness and a progressive decline in adult survival compared to controls.

Flies co-expressing two different insertions of Gem3GD17221 (P{GD17221}v49505 and P{GD17221}v49506) ubiquitously under the control of Scer\GAL4tub.PU, Scer\GAL41032.hx or Scer\GAL4da.PU are completely lethal at 25[o]C and 29[o]C. When expressed in muscle lineages under the control of Scer\GAL4Mef2.PR viable flies are seen at both 25[o]C and 29[o]C. However, when co-expressed with Dcr-2Scer\UAS.cDa these flies are almost completely lethal at 25[o]C, and no adults are recovered at 29[o]C. Complete lethality is seen at both 25[o]C and 29[o]C when Gem3GD17221 is expressed under the control of two further muscle drivers, Scer\GAL4c179 or Scer\GAL4how-24B. Expression of Gem3GD17221 in the somatic musculature under the control of Scer\GAL4G7 results in semi-viability at 25[o]C and almost complete lethality at 29[o]C. Pan-neuronal expression under the control of Scer\GAL4elav.PU or Scer\GAL4nSyb.PS results in viability at 25[o]C, but semi-lethality at 29[o]C. In the presence of Dcr-2Scer\UAS.cDa, flies expressed under the control of Scer\GAL4elav.PU at 29[o]C are completely lethal. Expression in motor neurons under the control of Scer\GAL4RapGAP1-OK6 is viable at 25[o]C and lethal at 29[o]C, and this is seen with or without the presence of Dcr-2Scer\UAS.cDa.

Co-expression of two different insertions of Gem3GD17221 (P{GD17221}v49505 and P{GD17221}v49506) in the eye under the control of Scer\GAL4GMR.PU does not lead to a rough eye phenotype.

Age-dependent progressive flight defects are seen when two different insertions of Gem3GD17221 (P{GD17221}v49505 and P{GD17221}v49506) are expressed under the control of Scer\GAL4elav.PU.

Flies co-expressing Gem3Scer\UAS.cCa, Gem3GD17221 and Gem3ΔN.Scer\UAS under the control of Scer\GAL4Mef2.PR are flight defective.

Adults expressing Gem3GD17221 under the control of the cardioblast-specific Scer\GAL4tin.CΔ4 driver show significantly reduced survival on day 6 after a shift to 29[o]C compared to control flies.

Adults expressing Gem3GD17221 under the control of Scer\GAL4elav.PLu (in the presence of Dcr-2Scer\UAS.cDa to increase the efficiency of RNAi) do not show a significant defect in avoidance of noxious temperature (46[o]C) compared to control flies.

Expression under the control of Scer\GAL4pnr-MD237 results in bristle morphology defects on the notum in 0% or 20-30% of the Scer\GAL4pnr-MD237 expression domain, depending on the insertion line used.

Expression under the control of Scer\GAL4pnr-MD237 results in the absence of 0% or 30-40% of the Scer\GAL4pnr-MD237-expressing area of the notum, depending on the insertion line used.
