Adult flies expressing RetGD45 (and UAS-Dicer2 to enhance RNAi efficiency) driven by Scer\GAL4esg-NP7397 combined with tub-Gal80[ts] (for temporal control) display significant decrease in midgut stem cell proliferation (assessed by number of pH3-positive cells) both during normal homeostasis and ageing and during chemical damage-induced regeneration. However, the number of stem cell per midgut as well as the ratios of their differentiated progeny are comparable to those observed in wild-type flies.

Adults expressing RetGD45 under the control of Scer\GAL4elav.PLu (in the presence of Dcr-2Scer\UAS.cDa to increase the efficiency of RNAi) do not show a significant defect in avoidance of noxious temperature (46[o]C) compared to control flies.

Expression of RetGD45 under the control of Scer\GAL4elav.PLu (in the presence of Dcr-2Scer\UAS.cDa to increase the efficiency of RNAi) results in viable flies or partial lethality, depending on the particular P{GD45} insertion line used.

Co-expression of RetGD45 can suppress the increased number of intestinal progenitor cells in mitosis characteristic for adult flies expressing any of the following: wgScer\UAS.cHa.T:Ivir\HA1, sggA81T.Scer\UAS or Src42ACA.Scer\UAS driven by Scer\GAL4esg-NP7397 (combined with tub-Gal80[ts] for temporal control) but fails to prevent the increased proliferation induced by expression of either upd1Scer\UAS.cUa or spis.Scer\UAS.
