The third instar larval wing disc dorsal compartment smaller size and apoptosis induced by the expression of pnutGD1512 from the second instar larval stage under the combined control of Scer\GAL4ap-md544 and Gal80[ts] are suppressed by the co-expression of Diap1UAS.cUa. pnutGD1512's apoptosis phenotype is not suppressed by p535A-1-4 homozygosity or by the co-expression of p53H159N.UAS.Ex; the pnutGD1512, p53H159N.UAS.Ex co-expression does not lead to the formation of wing disc tumors. The expression of pnutGD1512 also leads induces polyploidy/aneuploidy phenotype in the wing disc dorsal compartment, which is enhanced by the co-expression of either Diap1UAS.cUa or ykiUAS.cHa.
The co-expression of pnutGD1512 and ykiUAS.cHa under the control of either Scer\GAL4hh.PU or Scer\GAL4e22c leads to a much more severe enlargement of the wing disc posterior compartment as compared to the ykiUAS.cHa single expression condition; the Scer\GAL4hh.PU-driven co-expression also leads to some of the expressing cells moving and invading the wing disc anterior compartment.
The co-expression of pnutGD1512 and ykiUAS.cHa from the second instar larval stage under the combined control of Scer\GAL4ap-md544 and Gal80[ts] enhances the third instar larval wing disc dorsal compartment enlargement induced by the expression of ykiUAS.cHa from the second instar larval stage under the combined control of Scer\GAL4ap-md544 and Gal80[ts], even leading to the formation of tumors and to cell polarity defects (i.e. mislocalization of Discs large protein); this tumor phenotype is enhanced by the co-expression of bskDN.UAS. The pnutGD1512, ykiUAS.cHa co-expressing larvae fail to pupate and, instead, continue to grow to form giant larvae.
The expression of pnutGD1512 in combination with wtsKK101055, Ras85DV12.UAS, but not with scribHMS01993, dlg1HMS01954, l(2)glHMS01905, Syx7JF02436, Rab5GD10492, TSG101GD14295, PtenKK109278, Tsc1HMJ21477, CskHMS02277, agoHMS00111, EgfrUAS.cBa, Dl::NΔECN.UAS, dppUAS.cCa or wgUAS.cGa, from the second instar larval stage under the combined control of Scer\GAL4ap-md544 and Gal80[ts] also leads to the formation of tumors in the third instar larval wing disc.
The triple co-expression of pnutGD1512, Diap1UAS.cUa and stgUAS.Tag:HA from the second instar larval stage under the combined control of Scer\GAL4ap-md544 and Gal80[ts] leads to the significant enlargement of the third instar larval wing disc dorsal compartment, as compared to the pnutGD1512, Diap1UAS.cUa double expression condition.
The smaller third instar larval wing disc dorsal compartment and associated polyploidy/aneuploidy induced by the expression of pnutGD1512 from the second instar larval stage under the combined control of Scer\GAL4ap-md544 and Gal80[ts] are respectively suppressed and enhanced by the co-expression of BacA\p35UAS.cHa; this pnutGD1512, BacA\p35UAS.cHa co-expression leads to a severe decrease in the number of cells undergoing DNA replication (assessed by EdU labelling and phospho-H3 immunostaining), as compared to controls.
