Description

Imprecise excision of the P{EP} insertion in MlfEU2490, resulting in a 1.8kb deletion removing nucleotides +443 to +2258 of Mlf (coordinates relative to the transcription start site of isoforms A,B and D).

MlfΔC1 homozygous third instar larvae present a significant increase in both the number and size of circulating crystal cells (identified as Lozenge-positive cells, and which are still capable of melanization upon heat treatment), but other circulating cells have unaffected size, as compared to controls; MlfΔC1 heterozygotes also exhibit a (dominant) increase in the size, but not number, of crystal cells, as compared to controls.

Crystal cell maintenance is impaired in MlfΔ5-3/MlfΔC1 embryos.

64.0% of animals derived from a cross of homozygous females to homozygous males die as embryos, whereas 46.5% of animals derived from a cross of homozygous females to wild-type males die, indicating that that maternal effect lethality is partially rescued by a paternal Mlf+ copy. Embryos derived from a cross of heterozygous females to wild-type males have normal viability.

90.0% of animals derived from a cross of homozygous MlfΔ5-3 females to homozygous MlfΔC1 males die as embryos.

Homozygous adults lack interocellar bristles and the head macrochaetae are frequently bent or shortened. Ectopic venation is seen in the wing near longitudinal vein 2.

DnaJ-1A, MlfΔC1 double heterozygous third instar larvae present a significant increase in the number of circulating crystal cells (identified as Lozenge-positive cells), as compared to heterozygous or wild-type controls. DnaJ-1A, MlfΔC1 double heterozygous third instar larvae present a significant increase in the size of circulating crystal cells (identified as Lozenge-positive cells), as compared to each heterozygous or wild-type controls.

The increased number and size of circulating crystal cells observed in MlfΔC1 homozygous third instar larvae is suppressed by the expression of lzScer\UAS.cBa under the control of Scer\GAL4lz-gal4 or by N55e11 heterozygosity.

Xenogenetic Interactions
Statement
Reference

Expression of Hsap\MLF1Scer\UAS.cMa under the control of Scer\GAL4GMR.PF almost completely rescues the macrochaete and wing phenotypes seen in MlfΔC1 homozygotes. The reduction in life span that is seen in flies expressing Hsap\ATXN3tr.Q78.Scer\UAS.T:Ivir\HA1 under the control of Scer\GAL4elav.Switch.PO in the presence of RU486 is not modified by the addition of MlfΔC1/+.

Expression of MlfScer\UAS.cFa under the control of Scer\GAL4lz-gal4 rescues the crystal cell phenotype of MlfΔ5-3/MlfΔC1 mutants.
