Description

Imprecise excision of the P{EP}EP561 insertion (located 101bp 5' of PSR), resulting in a deletion that removes the sequence encoding the first 210 amino acids of PSR. A portion of the insertion remains.

Comment:

Deletion resulting from the imprecise excision of the progentor insertion which extends from the insertion site and removes 627 bases of coding sequence from the initiator ATG. Some P-element.sequences remain.

Detailed Description
Statement

PSRFM1 mutants are viable and fertile and do not display any developmental timing defects. PSRFM1 enhances the variegated phenotype of the w locus in the In(1)wm4 background but has no effect on the variegation in the Sb locus when combined with T(2;3)SbV.

Homozygous adults are viable and fertile with no obvious morphological defects.

Homozygous embryos show a similar number and pattern of clustered apoptotic cells (assayed using acridine orange staining) as wild-type embryos. Macrophages in homozygous embryos contain many engulfed apoptotic nuclei, as in wild-type embryos.

Mutant third larval instar eye discs show no detectable alteration in apoptosis (as detected by TUNEL staining).

At 40 hours after puparium formation (APF), pupal eye development is normal in mutants and a characteristic hexagonal lattice of interommatidial cells is seen. However, by 50 hours APF, a loss of interommatidial cells is seen, which results in ommatidial fusions. Apoptotic cells are detected by TUNEL staining at 49 hours APF in the mutant eye disc, but not in the wild-type controls.

Additional Comments
Genetic Interactions
Statement
Reference

Combination with a single copy of PSRFM1 does not modify the frequency of eye tissue to antenna-like malformed structures transformation in adult flies expressing TAF1BdsRNA.Scer\UAS under the control of Scer\GAL4ey.200.Exel nor the massive eye overgrowth in flies expressing ykiS168A.Scer\UAS.T:Avic\GFP-EGFP,T:Ivir\HA1 with Scer\GAL4GMR.PF and while there is tendency towards worsening the disrupted wing vein phenotype of flies expressing smo5A.Scer\UAS.T:Zzzz\FLAG driven by Scer\GAL4C-765, the effect is not statistically significant.

The rough eye phenotype caused by rprGMR.PW, grimGMR.PC or WGMR.PG is enhanced by one copy of PSRFM1 and is further enhanced by two copies of PSRFM1.
