Expression of Npc1aKK101077 under the control of Scer\GAL4phm.PO (using tub-Gal80[ts] to limit the time of expression) results in accumulation of lipid droplets in the prothoracic gland cells of third instar larvae as well as trafficking defects as suggested by enlarged Rab7 positive vesicles and Lamp1 marker accumulations.
Without the tub-Gal80[ts] limiting the onset of expression the animals show developmental arrest in first larval instar.
Expression of Npc1aKK101077 RNAi under the Scer\GAL4P0206 driver results in a significant delay in pupariation and increased pupal size. Third instar larvae show accumulations of lipid droplets in prothoracic gland cells.
